Ovation® RNA-Seq System V2

Our latest generation Ovation RNA-Seq System V2 provides a fast and simple method for preparing amplified cDNA from total RNA for RNA-Seq applications, now with enhanced transcript coverage and uniform distribution of sequencing reads. Amplification is initiated at the 3' end as well as randomly throughout the whole transcriptome in the sample. This feature makes the Ovation RNA-Seq System V2 ideal for amplification prior to Next Generation Sequencing, as it allows amplification of both mRNA and non-polyadenylated transcripts.

Together with the streamlined workflow, the purified SPIA cDNA can be ligated directly into a sequencing library using an Encore NGS Library System (Part Nos. 300, 301 and 302) or other suitable library construction methods from leading NGS platform providers.

The Ovation RNA-Seq System V2 is enabled by Ribo-SPIA technology, a rapid, simple and sensitive RNA amplification process developed by NuGEN. Using Ribo-SPIA technology and starting with as little as 500 pg total RNA, microgram quantities of cDNA can be prepared in approximately 4.5 hours.

The Ovation RNA-Seq System V2 (Part No. 7102) provides optimized reagent mixes and a protocol to process 8 or 32 total RNA samples, and is also available as an automation solution for processing 96 samples.

NOTE: With the release of Ovation RNA-Seq System V2 all sizes of Ovation RNA-Seq System (Part No. 7100) have been discontinued.

RNA-Seq Read Coverage of three different human transcripts: 2 ng of total RNA from MAQC samples (Brain or UHR) was amplified using either the Ovation RNA-Seq System V2 (Part No. 7102) or Ovation RNA-Seq System (Part No. 7100) and libraries constructed using the Encore NGS Multiplex System I. Single-read sequencing results were obtained using the Illumina Genome Analyzer IIx platform. Sequencing read coverage plots are shown for Beta Actin (top), GABA Receptor Associated Protein (middle), and GAPDH (bottom).
